中枢神経系の炎症は神経ミトコンドリアの機能を損なう可能性があり、一般的な神経炎症性疾患である多発性硬化症(MS)における軸索変性の一因となります。 今回我々は、細胞型特異的なミトコンドリアプロテオミクスと生体内バイオセンサーイメージングを組み合わせて、炎症が神経ミトコンドリアの分子組成と機能的能力をどのように変化させるかを詳しく分析する。 われわれは、マウス脊髄の神経炎症性病変が、ミトコンドリア酸化やカルシウム過負荷に先立って、広範かつ持続的な軸索ATP欠乏を引き起こすことを示す。 この軸索エネルギー欠乏は、電子伝達系機能の障害だけでなく、上流のトリカルボン酸(TCA)サイクル酵素の不均衡にも関連しており、実験モデルやMS病変では、重要な律速酵素を含むいくつかの酵素が神経ミトコンドリアで枯渇している。 注目すべきことに、個々のTCA酵素のウイルス過剰発現は、神経炎症性病変における軸索エネルギー欠損を改善することができ、MSにおけるTCAサイクル機能不全が治療で修正できる可能性があることを示唆している。
MS は、中枢神経系 (CNS) の炎症がミエリンの喪失と進行性の神経変性を引き起こす一般的な神経疾患です。 MS 患者の長期障害に対するこのような神経変性プロセスの重要性は十分に確立されています 1,2,3 が、CNS の炎症と神経変性の間の機構的な関連性はまだ解明されていません。 MS患者および対応する動物モデルから得られた新たな証拠は、神経ミトコンドリアが炎症シグナルを神経変性の結果に導く重要なハブである可能性を示しています。 この概念は、神経エネルギーの恒常性におけるミトコンドリアの本質的な機能 4 だけでなく、(1) 損傷したミトコンドリアが実験的およびヒトの神経炎症性病変に位置する軸索に蓄積することを示した以前の研究によっても裏付けられています 5、6、7。 (2) 神経細胞のミトコンドリアは疾患の経過とともに DNA 欠失を獲得します 8。 (3) このような炎症誘発性のミトコンドリア損傷は、電子伝達鎖 (ETC) の機能を損なうことが考えられます 2,9。
したがって、ミトコンドリアの損傷は、初期の MS の高度に炎症を起こした病変ですでに始まっており、その後、病気の経過とともにさらに増幅する可能性があります。 このプロセスは、炎症と神経変性の間だけでなく、初期の再発寛解とその後の進行性病状の間の関連性も提供すると考えられます2,9。 これらの洞察の結果、ミトコンドリアは、MS の疾患経過全体を通じて神経変性を防ぐための有望な治療標的として浮上しました。 残念ながら、これまでのところ、このような戦略は、少なくとも大規模な臨床試験においては、確実な臨床上の利点を提供できていません10。 この失敗の根底にある理由の 1 つは、神経炎症性病変においてミトコンドリアの分子機構と機能的能力がどのように損なわれるのかについての理解がまだ不完全であることです。 さらに、これまでのほとんどの研究は ETC 障害に焦点を当てている 8,11 が、根底にある高分子複合体の複雑な構造とその遺伝学を考慮すると、ETC 障害を治療の標的とするのは難しい可能性があります。
今回我々は、マウスMSモデル(実験的自己免疫性脳脊髄炎;EAE)におけるニューロンミトコンドリアの選択的ex vivoプロテオーム解析と組み合わせた新しいin vivoイメージング戦略を適用し、炎症誘発性ミトコンドリア病理の分子基盤と機能的影響を調査する。 我々は、広範な軸索ATP欠乏が急性神経炎症病変で始まり、慢性疾患段階まで持続することを明らかにした。 これらの生体エネルギーの欠乏は、ミトコンドリアの酸化還元またはカルシウム恒常性異常に先行します。 急性および慢性のEAE脊髄から単離された神経ミトコンドリアの選択的MitoTagベースのプロテオーム解析では、代わりに重要なTCAサイクル酵素の不均衡が明らかになり、イソクエン酸デヒドロゲナーゼ3(Idh3)やリンゴ酸デヒドロゲナーゼ2(Mdh2)などのいくつかの酵素が顕著に減少していることが判明した。 注目すべきことに、神経ミトコンドリアにおけるこれらの重要なTCAサイクル酵素の枯渇は、ヒトのMS病変でも明らかです。 最後に、Idh3 または Mdh2 の触媒サブユニットを過剰発現するウイルス遺伝子治療が、神経炎症性病変における軸索 ATP 欠損を部分的に回復できることを示します。 したがって、私たちの研究は、神経炎症に応答して神経ミトコンドリアの分子組成がどのように変化するかについての洗練された理解を提供します。 我々は、TCAサイクル酵素の枯渇が神経炎症性病変で起こる軸索エネルギー危機の重要なメディエーターであることを特定し、したがって治療介入の潜在的な標的を定義する。
